A. Transgenic Facility Overview A transgenic and knockout mouse facility is available in the Research wing at Sunnybrook and Women's. Both pronuclear injections and ES cell<-> embryo aggregations are available on a fee-for-service basis. Questions can be directed to Dr. Corrinne Lobe (480-6100 x3382, corrinne.lobe@sw.ca). Investigators making use of the service are required to have their animal protocols approved by the Animal Care Committee. For pronuclear injection of transgenes, submit a completed order form. A protocol for insert purification and a date for injection will then be set. Two weeks before the injection date, you will need to provide the purified insert and a gel photo of an aliquot of the insert. Following microinjection, mice will be housed until weaning age (3 weeks) in the transgenic facility. When pups are 2 weeks old, we can assist the researcher to do tail biopsies and ear punches for genotyping. The genotyping can be done by the facility for an additional fee. For internal clients, transgenic pups will be transferred to the investigator's mouse colony at 3 weeks. For external clients, mice will be shipped at 4 weeks. For ES cell<-> embryo aggregations submit a completed order form. Two vials of the ES cell line to be aggregated will be required. We will have all lines tested for pathogens before aggregation. When the ES cell lines are verified for pathogen-free status, a date for aggregations will be set. Following ES cell aggregation, mice will be housed until weaning age. At 3 weeks, chimeras will be transferred to the investigator's mouse colony for internal clients, and for external clients chimeras will be shipped at 4 weeks. If space permits, chimeras may be tested for germline transmission in the facility, but cost of housing beyond weaning age will be billed to the client. |

Tissue culture is for thawing and expanding ES lines and MAP testing in preparation for aggregation experiments. Up to 3 ES lines can be done for the $500 fee. Genotyping - Frozen tail biopsies $50. Genomic
tail DNA in ethanol $200. Germline testing of chimeras can be done if space permits and for the additional housing fee indicated (requires an additional 13 weeks post-weaning housing). All fees are in Canadian dollars and must be paid in advance, i.e. before shipping. Cheques should be made out to Sunnybrook & Women's College Health Science Centre and mailed to C. Lobe, SWCHSC, S-218, 2075 Bayview Ave, Toronto, ON, Canada M4N 3M5. Please indicate on the request form if an invoice is required. |
